The syndrome affects multiple systems with motor and non-motor features. In the past decade, many genes and specific mutations contributing to disease risk have been identified. In the Arab-Berber population of Tunisia in North Africa the burden of genetically-defined PD is especially high. In this project the NINDS Udall Center at MCJ and the National Institute of Neurology in Tunis aim to share clinical, genetic, pathological and scientific expertise, to work towards the common goal of defining the natural history of PD, while building research capacity in Tunisia. Our proposal has five main aims: 1) To build database infrastructure and informatic resources (with the support of Zaah Technologies); 2) Clinical assessment of parkinsonism in research subjects; 3) Clinical assessment of non-motor disability; 4) Assessment of pedigrees with familial parkinsonism, and; 5) Genetic assessment of known and novel genes for parkinsonism. Together we intend to foster longitudinal clinical and genetic research that was initiated by GlaxoSmithKline, but is no longer supported. Research capacity, resources and expertise in Tunisia at the National Institute of Neurology are to be building about a pragmatic need, developing ocal expertise and resources. Nevertheless, the insights gained in PD are likely to be of universal benefit. The training provided and infrastructure developed in Tunis may also facilitate research in other prevalent neurological disorders. PUBLIC HEALTH RELEVANCE: The work proposed is a natural history study of Parkinson's disease (PD). Our objective is to: 1) build research capacity, resources and expertise in the National Institute of Neurology, Tunis, and; 2) develop cross- sectional research initiated by GlaxoSmithKline into a collaborative, longitudinal study of the natural history of PD, and; 3) forge a close collaboration between a NINDS Udall Center, the Departments of Neurology and Neuroscience at MCJ with the National Institute of Neurology in Tunis. | CITY OF JACKSONVILLE | $73,981.00 | Grant | 16.804 - Recovery Act - Justice Assistance Grants - Localities The City of Jacksonville Beach is an eight and one-half square mile, incorporated city within Duval County, Florida. The city has 21,500 full-time residents and the county has approximately to 750,000 residents. The city has over 2,500 businesses, crowded roadways, and a downtown entertainment district that attracts large numbers of visitors. The daytime population swells to 50,000 visitors and residents with crowds reaching 75,000 on days between March and October. The police department responded to over 55,000 calls for service last year with a mere 61 sworn officers. The police department faces unique challenges because of these factors. Because of unknown funding levels in the future, The Jacksonville Beach Police Department decided to upgrade equipment in existing programs or areas of need. We decided against starting new programs or hiring additional personnel with the grant funds because of this. This funding will allow the department to upgrade existing radio equipment to better serve the residents and the hundreds of thousands of visitors from all over Northeast, Florida. This communications upgrade will directly benefit all public safety organizations in Jacksonville Beach and will be crucial in countywide communications and interoperability during emergency operations such as hurricanes or during event operations such as large public gatherings. The federal government suggests all public safety agencies become P-25 compliant over the next few years. The grant will be used as partial funding to upgrade the current City of Jacksonville Beach 800 MHz radio system from an analogue system to a digital system. Specifically, five repeaters at the tower site will be replaced as well as software that will allow public safety (911 dispatch, police, and fire) to utilize digital talk-groups. Other communication equipment upgrades are also needed. In addition it will allow the police department to encrypt their current operating channels and move the entire radio system closer to a P-25 or regional system. This grant will allow some of these upgrades to occur and will represent Phase 1 of the communications system transition. Future funding for the completion of the transition will be taken from the city radio fund and other revenue streams. Once the application is approved we will receive quotes and sole source pricing where applicable and in accordance with the city purchasing policy. The equipment will be purchased immediately after the appropriate approval process. The strategy is to expend the funds expeditiously, within the specified grant period, to maximize benefit to the department and the citizens who live and visit our community. Considerable data links vasopeptidases, including enothelin-converting enzymes (ECE)-1, ECE-2, neprilysin (NEP), and angiotensin-converting enzyme (ACE), to the degradation of the Alzheimer's amyloid B-peptide (AB). To date, only a fraction of AD cases can be attributed to known risk factors other than age, thus, it is quite possible that distinct subgroups of AD patients exist for whom decreased AB clearance is the precipitating factor for the disease. Many factors, such as genetic mutation, transcriptional alteration, or even pharmacological manipulation may decrease the activity of AB-degrading proteases in the brain. In our parent R01 grant, R01NS048554, we proposed to evaluate whether vasopeptidase deficiency affects AB levels in gene knockout mouse models, and in mice treated with clinically-relevant vasopeptidase inhibitors. Our data confirms that ECEs and NEP contribute to AB degradation in vivo, but indicates that ACE plays a more limited role in regulating AB levels. While decreased AB degradation increases the concentration of the more limited role in regulating AB levels. While decreased AB degradation increases the concentration of the peptide, the converse is also true; increased activity of any of the AB degrading enzymes lowers AB levels and can potentially protect against AD. Recently, a single culeotide polymorphism (SNP) in the promoter region of the ECE1 gene has been identified and reported to be associated with risk for late-onset AD (LOAD). This variant, C-338A, creates a transcription factor binding site, which suggests that it results in increased transcription of ECE. Consistent with the hypothesis that increased ECE activity could decrease AB levels, individuals homozygous for the C-338A variant were found to be at significantly lower risk for LOAD. These limited findings are the only published studies documenting a genetic association with an AB degrading enzyme and AD, in which the functional effects of the protein are altered by increasing mRNA levels. | MAYO CLINIC JACKSONVILLE | $430,738.00 | Grant | Trans-NIH Recovery Act Research Support The long-term goals of this competing renewal are to elucidate protein kinase C (PKC) signaling mechanisms that contribute to cancer and translate these mechanistic insights into better prognostic and treatment strategies. During the current funding period, we identified PKC? as a human oncogene in non-small cell lung cancer (NSCLC), the number one cancer killer in the United States. PKC? drives anchorage-independent growth and invasion in NSCLC cells by forming a protein-protein complex with the adaptor molecule Par6. The PKC?-Par6 complex activates the Rho family GTPase Rac1 to induce expression of matrix metalloproteinase10 (MMP10). MMP10 is a critical effector of the PKC9-Par6-Rac1 signaling axis and is required for anchorage-independent growth and invasion in vitro. These mechanistic studies informed the design of a rational drug screening strategy which identified the first PKC?-targeted drug, aurothiomalate (ATM) which is currently in Phase I clinical trials for treatment of lung cancer. In this application, we will address three major aspects of oncogenic PKC? signaling. First, we will determine how PKC? activates Rac1. Preliminary data indicate that the Rac-GEF Ect2 regulates Rac1, is required for NSCLC cell transformation, associates with the oncogenic PKC?-Par6 complex and is phosphorylated by PKC?. In Specific Aim 1 we will test the hypothesis that Ect2 is a critical effector of oncogenic PKC? and that PKC?-mediated phosphorylation regulates the oncogenic activity of Ect2. Completion of this aim will provide a novel functional link between Ect2 and PKC?. Second, we will assess whether the oncogenic PKC? signaling axis we defined in NSCLC cells in vitro is required for NSCLC tumor growth and metastasis in vivo. In Specific Aim 2, we will test the hypothesis that the PKC?-Par6-Ect2-Rac1-MMP10 signaling axis is necessary for NSCLC tumor growth and metastasis in vivo. We will establish novel orthotopic NSCLC tumor models to assess the role of key components of this signaling axis in vivo. Third, we will determine whether MMP10 is required for lung carcinogenesis in vivo. Preliminary data demonstrate that PKC? is required for lung carcinogenesis induced by oncogenic K-ras in vivo. In Specific Aim 3 we will test the hypothesis that MMP10 is a critical effector of PKC? in K-ras-mediated lung carcinogenesis in vivo. We will establish novel transgenic mouse models to assess whether MMP10 can drive spontaneous lung carcinogenesis, exacerbate K-ras-mediated lung carcinogenesis and/or restore K-ras-mediated lung carcinogenesis in mice lacking PKC?. Completion of these aims will provide critical new insight into oncogenic PKC? signaling, establish the role of Ect2 in NSCLC, assess the importance of oncogenic PKC? signaling in relevant preclinical NSCLC models in vivo, and determine if MMP10 is a critical mediator of lung carcinogenesis in vivo. These studies will likely reveal Ect2 and MMP10 as novel therapeutic targets for development of therapeutics for treatment of lung cancer. PUBLIC HEALTH RELEVANCE: Lung cancer is the number one cause of cancer death in the United States. Protein kinase C? (PKC?) is an oncogene, prognostic marker and therapeutic target in lung cancer. | MAYO CLINIC JACKSONVILLE | $152,867.00 | Grant | Trans-NIH Recovery Act Research Support Using gene array analysis of patient matched normal renal and clear cell renal cell carcinoma (ccRCC) tissues, our original observation showed loss of the type III TGFbeta receptor (TBR3) in patients diagnosed with ccRCC. Based upon this observation, we embarked upon elucidation of the role and regulation of TBR3 in normal primary renal cells (NREs) and its loss in ccRCC cell lines. TBR3 had previously not been viewed as biologically important since it possesses a short cytoplasmic domain and binds all three TGFbeta isoforms; it was thought only to enhance TBR2 signaling by passing off TGFbetas to TBR2. We were one of the first to show that TBR3 plays an important role in cancer as a tumor suppressor. Since then, loss of TBR3 has been identified in breast, prostate, pancreas, lung as well as ovarian cancers and linked to tumor suppressor functions. Using patient matched NREs and ccRCC cell lines in clture and animal models, we have since made seminal discoveries that include: 1) TBR3, when re-expressed in ccRCC cells, induces apoptotic cell death via activation of phosphorylated p38 MAPK, 2) TBR3 signaling is TGFbeta- and TBR2-indevependent requiring only the cytoplasmic tail of TBR3 for signaling, 3) GATA3 is discovered as the first known positive transcriptional factor upregulating human TBR3 mRNA and 4) GATA3 is epignetically silenced in ccRCC while TBR3 is not: Thus we have discovered a new signaling pathway linking GATA3 to apoptosis: GATA3->TBR3->pp38 MAPK->apoptosis. GATA3 is a key transcription factor regulating renal development and differentiation and may be linked to renal carcinogenesis via TBR3 loss. Based on these novel discoveries, we propose to explore in detail the mechanism of and structure: function of the TBR3 cytoplasmic domain in activating p38 MAPK leading to apoptosis. Methylation and HDAC inhibitors are promising anticancer drugs. We discovered that when combined, they caused massive cell death and will test whether TBR3 mediates this effect. Lastly, we identified a negative regulatory factor binding this region. The combined results have molecular implications in renal carcinogenesis and treatment for ccRCC patients.
