The purpose of the subject research is to delineate mechanisms ofLIN28 regulation in the context of reprogramming and metabolism. Dr. Daley's research generally focuses on the pathways that drive blood development and cancer. His laboratory co-discovered the biochemical connection between LIN28 and let-7, and his laboratory has generated several murine models demonstrating the potent oncogenic effect ofLIN28 and its suppression by let-7. His academic research is distinct from the Company's aims in that he explores the biology of the pathway and collaborates with other labs, who have performed pilot chemical screens to identify probe compounds that inhibit the LIN28/let-7 pathway. The company will pursue high throughput screening on an industrial scale, seeking hit compounds and then applying medicinal chemistry to optimize chemical properties to produce drug-like compounds. Dr. Daley, as the PI of the study will be involved in all aspects of the research. As such, the outcome of the research has the potential affect the Company.

LIN28/let-7 MECHANISMS IN REPROGRAMMING AND METABOLISM

This project investigates the function of a recently described pathway involving the LIN28 RNA binding protein and one of its key targets, the let-7 family of microRNAs. LIN28/let-7 has been implicated in several aspects of human physiology and disease, including the regulation of growth, puberty, metabolism, cancer, and diabetes. Moreover, LIN28 is one of the factors that facilitates the resetting of specialized adult cells back to a state resembling embryonic stem cells, a process termed reprogramming. This application will investigate the mechanism by which LIN28/let-7 influences cell proliferation and metabolism, and will use reprogramming as an accessible in vitro model system to dissect the structure-function relationships of LIN28B and its targets. Insights into how LIN28 is regulated and the impact on its many targets may point towards improved methods to generate stem cells and novel therapeutic interventions in conditions like cancer and diabetes
