This proposal applies rigorously defined genetically engineered mouse models of pancreas cancer to translate therapeutic regimens that target both the stromal and epithelial compartments of the disease. Dr. Hingorani’s previous work defined a strategy to administering a long-acting, PEGylated formulation of hyaluronidase (PEGPH20) in order to degrade intratumoral hyaluronan (HA), release the complexed water, reduce interstitial fluid pressure and allow collapsed vessels to re-expand and re-perfuse to increase penetration of subsequently administered agents. This approach is currently being tested in Phase 3 clinical trials. This current R01-funded research pursues a series of complementary investigations to rigorously investigate the role of hyaluronan (HA) in promoting the pathogenesis and aggressive biology of PDA (pancreatic ductal adenocarcinoma). The aims proposed in this 5-year project will involve administration of PEGPH20 in a variety of genetically engineered mouse models of pancreas cancer, using a variety of therapeutic strategies including PEGPH20 monotherapy as well as PEGH20 in combination with other therapeutic agents and approaches. PEGPH20 is provided free of charge from Halozyme Therapeutics.

PROJECT NARRATIVE Pancreas cancer kills every patient it afflicts and is now the fourth leading cause of cancer-related deaths in this country with a rising incidence and unabated mortality. We have developed genetically engineered animal models of pancreas cancer that have exposed unique points of vulnerability for the development of new treatments. We propose a systematic program of investigation to target distinct components of the stromal reaction in pancreas cancer as the basis for new treatment strategies against this highly resistant disease.
