Dr. Mostoslavsky is a co-founder of Galilei Biosciences, Inc. with equity in the private company. Galilei Biosciences Inc. seeks to develop activators of the mammalian SIRT6 protein. The research project tests whether SIRT6 regulates activation of metabolic and developmental genes through a novel, previously unrecognized mechanism of action, which may inform treatment against developmental diseases, metabolic diseases and cancer. The results of these research projects could therefore be of interest to the company. The Partners Committee on Outside Activities review panel evaluated the financial interest in connection with the research projects and determined that, based on the close connection between the company's interests and the research, the financial interest could directly and significantly affect the design, conduct, or reporting of the research.

Project Narrative Proper regulation of gene expression represents one of the most important characteristics of appropriate cell function; each one of our cells, despite carrying all the genetic information in its DNA, expresses only a subset of specific genes in order to maintain its cell fate, differentiation stage, and growing characteristics. At the molecular level, we are still trying to understand how expression of specific genes is fine-tuned and specifically coordinated; our laboratory has identified a protein, called SIRT6, that functions as a modulator of chromatin, a highly dynamic structure that compacts DNA, in turn acting as a key regulator of embryonic development, metabolism and cancer. In this proposal, we will use biochemical and biological approaches, as well as genetically engineered cells and induced pluripotent human cells from patients with mutations in SIRT6 to test the hypothesis that SIRT6 regulates activation of metabolic and developmental genes through a novel, previously unrecognized mechanism of action.
