Enhancing High-Intensity Focused Ultrasound for Tumor Ablation with Phase-Changing Nanoagents

High intensity focused ultrasound (HIFU) provides a non-invasive and inexpensive technique for treating tumors. With this technique, ultrasound waves are delivered to the tumor in order to rapidly heat the diseased tissue, or ablate the tissue, and cause cell-death. There are concerns with HIFU regarding heating healthy tissue along with the diseased. In addition, there are concerns that to implement HIFU clinically would require large amounts of ultrasound energy and long surgery times. Our lab aims to show that a developing ultrasound contrast agent, known as phase-changing nanoagents (PCNAs), can minimize the heating of undesired areas, while enhancing the efficacy of HIFU, thus decreasing surgical time and energy requirements. This award will allow the characterization of PCNA formulations based on the in vivo properties of circulation time and penetration ability within the tumor, both of which have not been explored well and hinder technology progression. These characteristics of PCNAs will directly aid in optimizing PCNA formation for use in a PCNA-mediated HIFU pre- clinical pilot study.
