Fatty-acid amide hydrolase targeted prodrugs for directing thyromimetics to the brain
Project Narrative The blood-brain barrier represents a major challenge in developing novel therapeutics for the central nervous system by significantly limiting biodistribution into the brain. There is significant interest in using the thyromimetic sobetirome for treatment in demyelinating disorders such as multiple sclerosis because thyroid hormone is known to regulate the development of oligodendrocytes. By designing prodrugs that target the brain resident protein fatty-acid amide hydrolase (FAAH), this research will test and validate a novel strategy for significantly increasing brain exposure of sobetirome while simultaneously limiting the peripheral effects of this drug.
