Dr. Madura holds equity interest and is the founder and acting CEO in a start-up company characterizing the ubiquitin/proteasome system to identify disease-specific biomarkers and targets for drug discovery in human diseases, and to develop diagnostic assays for use in the animal science industry. The company does not have any value at this time. Dr. Madura is also author of several of several patents and patents applications licensed by CellXplore Inc. from UMDNJ/Rutgers, on new methods for probing the ubiquitin/proteasome system. These patents do not have any IP value at this time. The current NIH grant is to characterize the yeast Rad23 protein which traffics multiubiquitinated proteins to the proteasome, and to investigate the role of the ubiquitin proteasome system in nucleotide excision repair. Since the subject matters of the grant, and of the company and patents utilize proteasome-based assays, the ICOI committee concluded that, while there might not be any direct overlap, sufficient relatedness existed to warrant the existence of the perception of a conflict of interest.

Functional Analysis of Rad23 Protein

The primary objective of the proposed studies is to investigate the role of protein degradation in DNA repair. Xeroderma pigmentosum and Cockayne Syndrome are well studied DNA repair defects for which there is no treatment or cure. Our studies could pave the way towards identification of candidate proteins and biochemical functions that can be targeted by therapeutic agents.
