A novel RNA-guided platform for sequence-specific cell reprogramming
The ability to reprogram cells with a novel identity and developmental fate opens new doors in studying human physiology and disease and raises the possibility of repairing malfunctioning tissues. However, most cellular reprogramming applications remain inefficient, partly due to our inability to manipulate large numbers of genes on a genome-wide scale and in a controlled and programmable manner. To address this major challenge, the proposed research aims to develop a novel genetic platform for cell reprogramming that is more efficient, more stable and safer, using generation of induced pluripotent stem (iPS) cells as a test bed, which is also applicable to other cell-based therapy strategies for disease treatment and tissue regeneration.
