Dr. Yeo is co-founder, chief executive officer and chief science officer of Locana. He holds (b)(4) equity interest valued at (b)(4). Locana is using CRISPER DNA editing tool to develop treatments for disease caused by RNA. This NIH subaward from University of Connecticut is for Dr Yeo’s lab to design and generate gene targeting constructs to enable the CRISPR-Case9 mediated tagging of endogenous genes; introduce these into K562 and HepG2 cell lines; perform eCLIP experiments to map the RNA binding sites of RNA binging proteins in these tagged cell lines, and develop portable software workflows for integrative analyses of all data generated in the community resource project.

OVERALL ? PROJECT NARRATIVE The ENCORE (Encyclopedia of RNA Elements) project produces physical resources and datasets that form the reference annotation for RNA binding protein-RNA interactions in the human genome, providing a standard for insights into co- and post- transcriptional regulation in the short run, and consequences of expressed genetic variation in the long term, contributing to fundamental knowledge to benefit public health.
