A Nanopatterned MEA-based Functional Phenotyping Assay for Drug-Induced Cardiotoxicity Screening

PROJECT NARRATIVE This proposal aims to develop a novel high-throughput in vitro assay for screening human stem cell-derived cardiomyocyte function in response to multiple maturation stimuli, and their subsequent reaction to treatment with known cardiotoxic compounds. We will screen multiple embryonic and induced stem cell lines in order to determine the level of functional variability present in cardiomyocytes derived from different sources. This information will enable the normalization of data from compound efficacy/toxicity studies using different cell sources, and provide effective baseline selection criteria for downstream preclinical drug-induced cardiotoxicity screening applications.
