A Multi-Omics Approach to Discover Metabolic Critical Quality Attributes for Cardiomyocyte Biomanufacturing
PROJECT NARRATIVE Human induced pluripotent stem cell (iPSC)-derived cardiomyocytes (CMs) offer the potential to functionally regenerate heart tissue in patients with heart disease and to evaluate drugs and other compounds for cardiotoxic effects, but iPSC-CMs lack adult phenotypes necessary to realize their promise. To overcome this roadblock, we propose an integrated metabolomic and proteomic profiling of both iPSC-CMs in vitro and cardiomyocytes in vivo to better understand metabolic transitions that occur during maturation and to predict multivariate critical quality attributes (CQAs, e.g. metabolites and metabolic enzymes) of maturation during iPSC-CM biomanufacturing. This study will provide greater insight into metabolic pathway utilization changes that occur during cardiomyocyte maturation, identify new CQAs for iPSC-CM quality, and develop tools to monitor these CQAs during cardiomyocyte biomanufacturing.
