PNA Nanoparticles for Gene Editing In Vivo
This application will develop novel chemically modified peptide nucleic acids (PNAs) for highly effective and non-toxic gene editing, delivered in vivo by polymer nanoparticles. PNAs are synthetic pieces of DNA that can bind to the chromosomes of cells in a targeted manner and stimulate recombination of the target site with single-stranded donor DNAs to produce gene editing, with extremely low off target effects compared to other approaches. We will test the novel PNAs for correction of a thalassemia-associated mutation in the ?-globin gene in mice using nanoparticle-mediated delivery via simple intravenous (IV) administration, a minimally invasive approach that could lead to a clinically applicable method for therapeutic gene editing.
