Role of Tau Conformations in Vascular Contribution to Cognitive Impairment and Dementia
Scientific evidence continues to support a major contribution of vascular insufficiency to the development of dementia such as Alzheimer's disease, but the underlying mechanisms remains elusive. We have recently discovered a novel toxic form of tau protein, cis P-tau that mediates the development of dementia associated with Alzheimer's disease and traumatic brain injury, but can be effectively blocked by cis mAb treatment. In this proposal, we have assembled a multidisciplinary team with all the requisite expertise to test our new hypothesis that similar toxic cis P-tau may connect initial vascular insults to later development of dementia, which would offer a novel early disease mechanism and a promising new therapeutic approach.
