Proteostasis Therapeutics, Inc.

This proposal aims to examine the basis of intertissue signaling in the regulation of organismal proteostasis, and specifically investigating how intertissue signaling affects the organismal response to proteotoxicity during aging, using a model organism, C. elegans. This proposal also looks to investigate how proteotoxic proteins Abeta, tau or polyQ (misfolding and/or aggregation of which are all associated with diseases) affect peripheral proteostasis, and whether the organismal proteostasis network (PN) can be restored through genetic and small molecule approaches. Proteostasis Therapeutics has the goal of developing novel therapeutics designed to characterize and pharmacologically control the proteostasis network in cells, and which focuses on protein aggregation diseases. While the work included in this proposal examines basic mechanisms relating to proteostasis, the similarity of these studies and the broad goals of Proteostasis Therapeutics suggest the potential for a perceived conflict of interest that should be managed.
Regulation of Peripheral Proteostasis
PROJECT NARRATIVE Among the most daunting diseases are those known as protein conformational diseases that cause neurodegeneration including Alzheimer's disease, Parkinson's disease, ALS and Huntington's disease. Each of these diseases and hundreds of others are due to protein aggregation and resulting tissue failure. This proposal is to address how different tissues communicate and coordinate the cellular response to aggregation and proteotoxicity.
