Based on Washington University’s Research COI Policy and the Procedures for Determining and Managing Research Financial Conflicts of Interests, Washington University determined Dr. Mahajan’s personal financial relationship with TechnoGenesys related to the above referenced research creates a financial conflict of interest due to the following reasons:

• Dr. Mahajan’s personal financial relationship is directly related to the research because this study is evaluating a technology licensed by TechnoGenesys.

• Because Dr. Mahajan owns equity in the company, he could directly benefit should the company’s equity value increase as a result of the study.

• Because Dr. Mahajan will earn royalties from the sale and distribution of the licensed technology, he could directly benefit, should the study impact the value of the intellectual property.

• Dr. Mahajan’s role, as described below, could directly affect the study.

While the committee determined financial conflicts of interest exist, they concluded sufficient mechanisms exist to ensure the objectivity and integrity of the research, based on the following reasons:

• The research is basic in nature and in its early stages.

• Dr. Mahajan is uniquely qualified by virtue of his expertise and experience, and the research could not be conducted as safely or effectively without his involvement.

• Additionally, a plan has been developed to mitigate the financial conflict of interest.

Molecular Mechanisms of Castration Resistant Prostate Cancer Recurrence & Therapeutic Strategies

We have discovered a novel signaling loop, wherein AR protein coordinates with the kinase ACK1 and facilitates modification of histone H4 leading to AR transcriptional activation. AR upon acetylation regulates ACK1 expression. To achieve therapeutic benefit from this new signaling event, we generated a novel ACK1 small molecule inhibitor (R)-9bMS that not only inhibits ACK1 activity and AR expression but also suppresses CRPC tumor growth in mice. In this proposal, we will perform detailed drug development, mechanistic, and preclinical therapeutic studies necessary to credential this treatment approach, with the long-term goal of ultimately advancing this therapy to the clinic.
