Based on Washington University’s Research COI Policy and the Procedures for Determining and Managing Research Financial Conflicts of Interests, Washington University determined Dr. Mahajan’s personal financial relationship with TechnoGenesys related to the above referenced research creates a financial conflict of interest due to the following reasons:
• Dr. Mahajan’s personal financial relationship is directly related to the research because this study is evaluating a technology licensed by TechnoGenesys.
• Because Dr. Mahajan owns equity in the company, he could directly benefit should the company’s equity value increase as a result of the study.
• Because Dr. Mahajan will earn royalties from the sale and distribution of the licensed technology, he could directly benefit, should the study impact the value of the intellectual property.
• Dr. Mahajan’s role, as described below, could directly affect the study.
While the committee determined financial conflicts of interest exist, they concluded sufficient mechanisms exist to ensure the objectivity and integrity of the research, based on the following reasons:
• The research is basic in nature and in its early stages.
• Dr. Mahajan is uniquely qualified by virtue of his expertise and experience, and the research could not be conducted as safely or effectively without his involvement.
• Additionally, a plan has been developed to mitigate the financial conflict of interest.
Targeting a Novel Epigenetic Signaling Nexus ACK1-pY88H4-AR/AR-V7 in Drug Resistant Metastatic Prostate Cancer
Project Narrative Presence of elevated AR mRNA levels in spite of prolonged androgen receptor (AR) antagonist treatment in castration resistant prostate cancer (CRPC) is a paradox that has mystified researchers. This proposal uncovers a novel mechanism of auto- regulation of AR transcription wherein AR protein coordinates functionally with ACK1 tyrosine kinase to modify chromatin within the AR gene. Significantly, these data have exposed an Achilles' heel for targeting new treatment strategies in CRPCs. Our preliminary data indicates that reversal of these epigenetic marks by a novel ACK1 inhibitor (R)-9bMS, that we have developed, opens up a new therapeutic option for this essentially incurable malignancy. This proposal will perform detailed mechanistic & in vivo studies to test efficacy of (R)-9b as novel epigenetic inhibitors that suppress both AR and AR-V7 transcription overcoming CRPC tumor growth.
